Palmitoylethanolamide (PEA or Palmidrol INN) is an aliamide with cannabimimetic effects. It is able to control mast cell reactivity and down-modulate mast cell degranulation, thus reducing the release of biological mediators with hystolesive (e.g. NO), hyperalgesic (e.g. NGF) and inflammatory (e.g. TNF) effects. On canine mast cells freshly isolated from skin biopsies, palmitoylethanolamide proved to reduce the release of histamine, TNF-alpha and PGD2. In an animal model of interstitial cystitis, palmitoylethanolamide was shown to restore the normal micturition threshold. All things considered, Palmitoylethanolamide may normalize urethra/bladder reactivity. Glucosamine hydrochloride (HCl) is the natural precursor of glycosaminoglycans (GAGs) and is well absorbed after oral administration. It enhances the synthesis of GAGs, thus helping the urothelial repair in the event of LUTD. Hesperidin is a natural flavonoid free from side effects. After oral administration, its active form (hesperitin) is detected in urine, where it exhibits beneficial antibacterial properties and protects vesicourethral cells against oxidative stress.
